    Tamoxifen diabetes

For estrogen-receptor positive breast cancer cases, tamoxifen has been the most important adjuvant hormonal therapy for the purpose of reducing recurrence rates and prolonging disease free survival. However, several side effects have been noticed, and fatty liver is one of the most common side effects among them. Since fatty liver is a common problem in the general population, we wanted to examine the effects of tamoxifen under pre-existing fatty liver conditions and evaluate the prevalence of tamoxifen-related impaired liver function. We recruited breast cancer cases at ages 20−70 years and divided them into tamoxifen or control groups. Personal information was collected, and fasting blood tests and abdominal ultrasound were performed. The changes of fatty liver degree between the initial and follow-up ultrasound were divided into five categories. Of the 406 enrolled participants, 266 were in the tamoxifen group and 140 were in the control group. Key clinical point: Prophylactic tamoxifen reduced breast cancers in vulnerable women, compared with placebo, but didn’t affect overall mortality. Major finding: Five years of tamoxifen treatment translated into a 30% decrease in the incidence of breast cancers among at-risk women, but there was no survival benefit at 20 years of follow-up. Data source: The IBIS-1 trial randomized more than 7,000 women to 5 years of either tamoxifen or placebo. Disclosures: The IBIS-I trial was supported by Cancer Research UK. Cuzick has received funding for other trials from Astra Zeneca and is a consultant for Astra Zeneca. The company also provided both the study drug and placebo. SAN ANTONIO – Five years of tamoxifen provided 20 years of breast cancer prevention to some at-risk women who took it prophylactically. However, their 20-year all-cause mortality was no different from those taking placebo (182 vs.

I did a little research on the Internet and do read there is a connection between diabetes and Tamoxifen.

Lorraine Lipscombe, scientist at Women’s College Research Institute and endocrinologist, reports surprising findings about a commonly used breast cancer medication. Her team found that in older breast cancer survivors, tamoxifen is associated with a significantly increased risk of diabetes, even after adjustment for other risk factors. “Tamoxifen is an important treatment for estrogen-receptor breast cancer that has been shown to improve breast cancer survival,” says Lipscombe. “We found that diabetes was 24 per cent higher in women who were using tamoxifen treatment, not with previous tamoxifen users. “This suggests that although current tamoxifen therapy may increase the risk of diabetes in people who are susceptible, this risk does not continue after the medication is discontinued.” The researchers suspect that the underlying mechanisms may be related to tamoxifen’s estrogen-inhibiting effect. Evidence suggests that estrogen plays a role in regulating blood glucose levels, and indeed post-menopausal women, whose estrogen levels are naturally lower, have a higher prevalence of diabetes, weight gain, insulin resistance and metabolic syndrome. Furthermore, post-menopausal estrogen replacement therapy has been shown to reduce the incidence of diabetes. About 1.5 years after taking Tamoxifen, I had a slightly elevated fasting blood glucose on an annual visit with my primary doctor. She wasn't concerned, but I was and started monitoring my blood sugar. It's always higher (and above the recommended 126) when I eat simple carbs. The study - conducted in nonhuman primates with brain structures and functions similar to those of humans - found that the antidepressant sertraline, a selective serotonin reuptake inhibitor (SSRI) marketed as Zoloft, significantly increased the volume of one brain region in depressed subjects but decreased the volume of two brain areas in non-depressed subjects. "These observations are important for human health because Zoloft is widely prescribed for a number of disorders other than depression," said Carol A. D., professor of pathology-comparative medicine at Wake Forest Baptist and lead author of the study, published in the current online issue of the journal Neuropharmacology. In the study, 41 middle-aged female monkeys were fed a diet formulated to replicate that consumed by many Americans for 18 months, during which time depressive behavior in the animals was recorded. Female monkeys were chosen for this study because depression is nearly twice as common in women as men and the use of antidepressants is most common in women ages 40 to 59. After the 18-month pre-study phase, the monkeys were divided into two groups balanced for body weight, body mass index and depressive behavior. For the next 18 months, 21 monkeys received sertraline in daily doses comparable to those taken by humans while a group of 20 received a placebo. This treatment regimen is analogous to a human taking an antidepressant for approximately five years.
